Academician Jiang Guibin's research group of the State Key Laboratory of Environmental Chemistry and Ecotoxicology, Research Center for Eco-Environmental Sciences, Chinese Academy of Sciences has made important progress in the estrogen effect of the new pollutant perfluoroiodide. Related research papers were recently published in the journal Environmental Health Perspectives.
Perfluoroiodine compounds are a new class of pollutants, and their toxic effects are blank. In this work, the estrogen effects of 12 perfluoroiodines were studied through cell proliferation experiments, MVLN experiments, and the expression of estrogen-responsive genes in MCF-7 breast cancer cells. The study found that iodinated perfluorinated compounds can induce cell proliferation, activate estrogen receptors in MVLN cells and induce the expression of TFF1 and EGR3 in MCF-7 cells, with estrogen effects. The estrogen receptor antagonist tetrahydroxytamoxifen combined with perfluoroiodine was exposed to MVLN cells, which further confirmed the effect of perfluoroiodide compounds on estrogen receptor activation. By comparing the estrogen effects of different perfluorinated compounds and perfluoroiodine, the study found that the estrogen effect of perfluoroiodine is related to the length of the perfluorocarbon chain, the degree of fluorine substitution, and iodine substitution. Its structure is obviously different from traditional estrogen compounds with phenolic hydroxyl structure. The mechanism of the estrogen effect of perfluoroiodine compounds is speculated, and its hydroxylated metabolite may be the target molecule for estrogen receptor activation.
This study first discovered and confirmed the estrogen effect of perfluoroiodine, explaining its structure-activity relationship, and providing an important basis for studying the endocrine disrupting effects of perfluorinated compounds.
