In angiosperms, scientists have discovered a protective structure for blunt terminal telomeres that is different from previous understandings.
The single-stranded telomere DNA protruding structure is considered to be an evolutionarily conserved basic structural element that plays an important role in protecting the ends of chromosomes. Previously it was thought that the formation of this DNA protruding structure at the telomere site was caused by the replication of the leader chain mechanism. This process involves a blunt end treatment after DNA replication whose mechanism is unclear.
Unexpectedly, this study found that angiosperms contain blunt ends and short (1-3 nucleotides) overhanging telomere structures containing G nucleosides, which can be stably retained in post-mitotic tissues. Thus revealing a new mechanism for protecting the ends of chromosomes.
The integrity of the blunt-end telomere depends on the Ku70 / 80 heterodimer, but not the capping protein on the other telomere, STN1. Strangely, the telomeres that lack the Ku protein are fully functional. These telomeres are cleaved by exonuclease 1, thereby promoting intrachromosomal recombination, which may help to form alternative chromosome end capping structures.
These data question the belief that telomeres require single-stranded DNA protrusions to form a functional capping structure, and prove that nature has flexible solutions to the protection of chromosome ends.
